INTENDED USE

Anti-HCV ELISA isan enzyme-linked immunosorbent assay (ELISA) for qualitativedetectionof antibodies to hepatitisCvirus inhumanserumor plasma. It is intended for screeningblooddonorsanddiagnosingpatients relatedtoinfectionwithhepatitisCvirus.

PROCEDURE

Reagentspreparation:

Allow the reagents to reach room temperature (18-30°C). Check theWash buffer concentratefor thepresenceofsaltcrystals. Ifcrystalshaveformed, resolubilizebywarmingat37°Cuntilcrystals dissolve.DilutetheWashbuffer (20X)asindicatedintheinstructionsforwashing.Usedistilledordeionizedwater andonlycleanvesselstodilutethebuffer.AllotherreagentsareREADYTOUSEASSUPPLIED.

Step1 Preparation:Mark threewellsasNegativecontrol (e.g.B1,C1,D1), twowellsasPositive control (e.g.E1,F1)andoneBlank(e.g.A1,neithersamplesnorHRP-Conjugateshouldbe added into theBlankwell). If theresultswill bedeterminedbyusingdualwavelengthplate reader, the requirement for useof Blankwell couldbeomitted.Useonlynumber of strips

Step2 requiredforthetest.

Step3 AddingDiluent:Add100μlofSpecimenDiluent intoeachwellexcept theBlank. AddingSample: Add 10μl of Positivecontrol, Negative control, andSpecimen into their respectivewells except theBlank. Note: Usea separatedisposal pipette tip for each

Step5 tappingtheplategently. Incubating:Covertheplatewiththeplatecoverandincubatefor30minutesat37°C. Washing:At theendof theincubation, removeanddiscardtheplatecover.Washeachwell5 timeswithdilutedWashBuffer.Each timeallowthemicrowells tosoak for30-60seconds

Step6 Afterthefinalwashingcycle,turndowntheplateontoblottingpaperorcleantowel,andtapitto

Step8 AddingHRP-Conjugate:Add100μlHRP-ConjugateintoeachwellexcepttheBlank. Incubating:Covertheplatewiththeplatecoverandincubatefor30minutesat37°C. Washing:At theendof theincubation, removeanddiscardtheplatecover.Washeachwell5 timeswithdilutedWashingbuffer.Eachtimeallowthemicrowellstosoakfor30-60seconds.
