Testing Principle
Safranin O is a basic dye, while fast green is an acidic dye. The basophilic cartilage matrix binds with safranin, appearing orange-red. The acidophilic subchondral bone and bone tissue bind with fast green, appearing blue-green or gray-green.

Stain Results

Cartilage matrix: Appears orange-red.

Subchondral bone, bone tissue, and collagen fibers: Appear blue-green or gray-green.

Red blood cells: Appear green.

Cell nuclei: Appear purplish-blue.
